Single-Tube Total RNA Extraction in About 15 Minutes

Recover intact total RNA from cell-based and low-input samples with RNAGEM. Built on the Exymes thermophilic proteinase and a simple buffer system, RNAGEM lyses cells with a single temperature change and delivers RT-qPCR-ready RNA in about 15 minutes, with no phenol, no chloroform, no chaotropic salts, and no spin columns or magnetic beads.

Designed for Cell-Based and Low-Input Samples

Recover RNA from a wide range of cultured and sorted material, including adherent and suspension cell cultures, cell pellets, cells stored in RNAlater, FACS-sorted populations, and laser capture microdissection samples. Yields are linear from around 10 to 100,000 cells, which makes RNAGEM well suited to single-cell work where every copy matters. The handbook also covers viral RNA from saliva and from swabs stored in transport media.

Complete Size Range, Straight to RT-qPCR

RNAGEM releases total nucleic acid, and the included DNase I removes genomic DNA to leave clean total RNA. Because there is no size-selection step, the full transcript range is retained, from ribosomal and messenger RNA down to small RNAs from approximately 18 nucleotides upwards. RT-PCR and RT-qPCR reagents can be added directly to the lysate with no further purification, so the kit fits neatly into 96-well and automated workflows.

A Fast, Hands-Off Workflow

Activate the proprietary proteinase at 75°C to lyse cells and destroy RNases, then inactivate at 95°C, all on a standard thermal cycler. The single-tube, closed process needs no mechanical disruption, no washes, and no transfers, which preserves RNA integrity, reduces contamination risk, and cuts plastic and hazardous waste. RNA-ready extract takes about 15 minutes, and the optional DNase digestion to remove genomic DNA adds only a few minutes more. One protocol scales from low-throughput bench work to high-throughput automated runs.
